FOXO4-DRI is a synthetic D-retro-inverso peptide derived from the FOXO4 sequence that engages the p53 binding interface within senescent cells. It is one of the original “senolytic” tool peptides, developed to selectively eliminate senescent cells.
FOXO4-DRI disrupts the FOXO4–p53 interaction in senescent cells, freeing p53 to translocate to mitochondria and trigger apoptosis selectively in cells under senescence-associated stress. Healthy cells lacking this FOXO4–p53 dependency are largely spared in preclinical models.
